南湖新聞網(wǎng)訊 近日,華中農(nóng)業(yè)大學(xué)水產(chǎn)學(xué)院魚類發(fā)育與遺傳育種團(tuán)隊(duì)梅潔教授課題組和武漢大學(xué)肖銳課題組合作研究成果發(fā)表,研究揭示了m6A讀取器Igf2bp3通過調(diào)節(jié)m6A修飾生殖質(zhì)基因的表達(dá)以實(shí)現(xiàn)斑馬魚生殖質(zhì)組裝的分子機(jī)制。
生殖質(zhì)是具有特定形態(tài)結(jié)構(gòu)的細(xì)胞質(zhì),包含卵母細(xì)胞遺傳的各種RNA和蛋白質(zhì)復(fù)合物,對生殖發(fā)育起到至關(guān)重要的作用。生殖質(zhì)組裝是將生殖質(zhì)RNA和蛋白招募到指定位置并進(jìn)行組裝的動態(tài)過程,決定了未來生殖細(xì)胞的命運(yùn)。大量研究表明在斑馬魚早期胚胎發(fā)育過程中,Bucky ball (Buc)通過調(diào)控生殖質(zhì)mRNA表達(dá)在生殖質(zhì)組裝過程中發(fā)揮重要功能,但其RNA調(diào)控機(jī)制還不清楚。
研究發(fā)現(xiàn)m6A讀取器Igf2bp3與Buc相互作用,并與之共定位于生殖質(zhì)中。與buc突變體表型相似,igf2bp3母源突變導(dǎo)致異常的生殖質(zhì)組裝和生殖細(xì)胞分化缺陷,該表型可通過注射igf2bp3 mRNA進(jìn)行部分挽救。進(jìn)一步研究發(fā)現(xiàn),Igf2bp3作為m6A識別蛋白通過結(jié)合m6A修飾的生殖質(zhì)相關(guān)mRNA來抑制其降解。
該研究揭示了Igf2bp3作為Buc的直接效應(yīng)蛋白,介導(dǎo)m6A修飾依賴的生殖質(zhì)相關(guān)mRNA的基因表達(dá)調(diào)控,并維持正常的生殖質(zhì)組裝(圖1)。該工作闡述了m6A修飾在魚類生殖質(zhì)組裝過程中的作用,為魚類生殖發(fā)育研究提供了參考。

Bucky ball (Buc) is involved in germ plasm (GP) assembly during early zebrafish development by regulating GP mRNA expression via an unknown mechanism. The present study demonstrates that an m6A reader Igf2bp3 interacts and colocalizes with Buc in the GP. Similar to the loss of Buc, the genetic deletion of maternal igf2bp3 in zebrafish leads to abnormal GP assembly and insufficient germ cell specification, which can be partially restored by the injection of igf2bp3 mRNA. Igf2bp3 binds to m6A-modified GP-organizer and GP mRNAs in an m6A-dependent manner and prevents their degradation. These findings indicate that the functions of Igf2bp3, a direct effector protein of Buc, in GP mRNA expression and GP assembly involve m6A-dependent regulation; these results emphasize a critical role of m6A modification in the process of GP assembly.
